Abstract(in English) The author describes newly conducted quantitative and sensitive microassays for uric acid (UA) and creatinine determination, using uricase-quinoneimine-dye and creatinine-deiminase-indophenol methods. This UA assay uses the catalyzed quinoneimine dye reaction for the determination of hydrogen peroxide. The procedure was adapted for the determination of UA after oxidization with uricase. The final product in this reaction, which is the maximum purple color of quinoneimine dye, required 5 min at 37℃ or 10 min at room temperature, and absorbance was measured at 570 nm using a microplate reader. This creatinine assay uses the catalyzed indophenol reaction for the determination of ammonia. The blue indophenol as the final product required 10 min at 60℃. We demonstrated that this convenient colorimetric microplate-based rapid assay can be applied to determine UA and creatinine as an evaluation of renal function in biological fluids, such as urine, serum or plasma, and spent dialysate, during hemodialysis. This rapid and reliable assay system is very useful as the primary screening of urinalysis. In this study, the author has developed the novel microassays to determine urea which reported previously, UA and creatinine as typical three test items for renal function evaluation.
