Simple and Rapid Renal Function Analysis Using Newly Conducted Urea Microassay

Abstract(in English) The author describes a newly conducted quantitative and sensitive microassay for urea determination using an urease-indophenol method. This assay uses the catalyzed indophenol reaction for the determination of ammonia. The procedure was adapted for the determination of urea after hydrolysis with urease. The final product in this reaction, whichis the maximum blue color of indophenol, required 5min at 60℃, and absorbance was measured at 570nm. We demonstrated that this convenient colorimetric microplate-based rapid assay can be applied to determine urea as an evaluation of renal function in biological fluids, such as urine, serum or plasma, and spent dialysate, during hemodialysis. This rapid and reliable assay system is very useful as the primary screening of urinalysis, and also useful to understand in renal function at practical training for students of clinical engineering.
