An Estimation of Cell Forces with Hierarchical Bayes Approach Considering Cell Morphology

Abstract(in English) To elucidate the migration mechanism of a biological cell, a mechanical force generated by the cell is measured in many biological studies. One of popular techniques is to measure the locations of fluorescent nano beads which are embedded in the surface of the culture substrate. A force is estimated from bead transitions. However, it is difficult inverse problem to estimate forces from the transitions because the number of possible force points is much larger than that of embedded beads due to physical limitations. To solve this problem, in this study we propose Bayesian force estimation by introducing priors of the cell morphology and the force directions, with the EM algorithm for parameter estimation. Our algorithm shows a significant improvement in estimating a force.
