Detection of change in mechanical properties of a single DNA molecule stretched by dual trap optical tweezers

Abstract(in English) When DNA is stretched by dual trap optical tweezers, we can obtain not only force but also local spring constant of a single DNA by calculating auto-correlation and cross correlation of the fluctuation of attached beads. To apply the correlation methods to detect DNA-protein interaction without labeling, we investigated the appropriate condition of experimental setup and its ability for detection. We varied laser power and local spring constant of a stretched DNA, k_, was obtained by using the correlation method at each laser power. The results show that appropriate spring constant of optical tweezers is 4-5 pN/μm for our setup, and we can detect the change of k_ with 0.4 pN/μm accuracy, with the time resolution of 1 s.
