Mathematical analysis of space-clamp problems in voltage-clamp recording

Abstract(in English) The measurement of synaptic current at the distal dendrite with the somatic voltage clamp has an error due to a failure of space clamp in the whole neuron. Williams & Mitchell quantified space-clamp errors by using the simultaneous recording from the soma and apical dendrite of rat cortical layer V pyramidal neurons. It has been reported that the electrotonic length of cortical layer V pyramidal neuron is about 1. Contrary to this, Williams & Mitchell showed the space-clamp error is larger than that of the electric cable with electrotonic length 1. Recent experimental and theoretical studies have suggested that the membrane resistivity of the distal apical dendrites of cortical layer V pyramidal neurons may be significantly lower than that of the proximal dendrites and the soma. Therefore, it is possible that the large space-clamp error is due to the low membrane resistivity at distal apical dendrites. In this paper, we formulate an electric behavior of the apical dendrite with the leaky distal part when clamping the somatic membrane potential as a finite passive cable equation with the fixed and leaky boundary conditions, and by employing Green function method we elucidate the effect of the low membrane resistivity at distal apical dendrites on the space-clamp error. The solutions for a cable with a leak at one end qualitatively reproduce the space-clamp errors measured by Williams & Mitchell, whereas those without the leak cannot reproduce them even its electrotonic length is adjusted.
