Study on the antimicrobial activity of Hinokitiol examined by colony counting method and intracellular ATP dose assay

Abstract(in English) In the previous study, we demonstrated that the small amounts of Hinokitiol which isolated from tree oil inhibited bacterial growth effectively. About 50μg/ml (=ppm) of Hinokitiol clearly suppressed either of Escherichia coli and Staphylococcus aureus growth when they were incubated in optimum growth condition like as broth medium, whereas about 100μg/ml (=ppm) of Hinokitiol administration showed bactericidal activity against employed bacteria in same growth condition. In this present study, we conducted the measurement of bacterial metabolic activity using intracellular ATP assay. About 50μg/ml (=ppm) of Hinokitiol did not affect the growth rate of E. coli and Sta. aureus when its were measured by colony forming examination, however, the ATP doses in both of bacterial strains were remarkably decreased by HK administration. This phenomenon indicated that small amount of HK cloud promote the VNC (Viable but not cultured) condition. These result suggested a newly possibility of HK utilization for infection control substance.
