Abstract(in English) | Alzheimer's disease (AD) has been studied for the changes of molecules such as amyloid β. Few studies have investigated the change of the whole signal transduction system, preventing the elucidation of the mechanism. We aimed to elucidate the structural features of human hippocampal signal transduction system and the rules and mechanisms which emerge in AD. We constructed AD specific signal transduction network as a graph using gene expression and weighted directed signal transduction pathway data of hippocampus, and evaluated the changes of the system by commonly used statistical indexes and k shortest path. As a result, although the network density, transitivity, average clustering coefficient and shortest path length in the AD network resembled those in the network, the nodes of which were removed randomly, the robustness differed in the specific input and output sets in the k shortest path analysis. Applying this model, we successfully identified the changes of robustness in human hippocampal signal transductions in AD due to the differential gene expressions. |
