Abstract(in English) | Polypeptide thin films have been prepared on gold surfaces by an all dry vacuum process by evaporating polymerization initiator followed by amino acid anhydride. The film growth process was observed by in-situ surface plasmon resonance (SPR). In the first stage, amino ethane thiol (AET) was deposited on gold surface as a polymerization initiator. Although conventional vapor deposition failed in accumulating AET films, growth of AET films was made possible by utilizing hot-filament assist in the course of AET evaporation. In the second stage, γ-benzyl-L-glutaminate N-carboxy anhydride (NCA) was evaporated on the AET films. The growth process of NCA film was dependent on the AET thickness. When the AET thickness was smaller than 15 nm, uniform thin polypeptide films grew in proportion to the deposition time. When the AET layer was thicker than 20 nm, on the other hand, the polypeptide grew in two stages; uniform planar growth up to 50 nm, followed by rapid growth of instable film. It was concluded that hot-filament-assisted deposition of thin AET layer followed by vapor deposition of NCA enables to form uniform polypeptide thin films by an all dry physical vapor deposition process. |
