Development of a droplet generator for single-molecule fluorescence imaging

Abstract(in English) We have a plan to used solvent-free air-substrate interfaces on which target molecules are detected and identified. To do so, we developed a droplet generator to arrange droplets containing target molecules on a substrate surface. We arranged droplets of a rhodamine B aqueous solution on a silicon substrate, and found that the droplets were arranged at intervals of 50 μm by imaging fluorescent spots with a CCD camera equipped with an optical microscope after droplets dried up. The average volume of the droplets was 2.2pl. However, the reproducibility of droplet volume was poor because of deterioration in the electrode with time. The electrode was a deposited metal on a glass micropipette in the droplet geneerator. After exchanging the deposited metal for the tungsten wire inserted into the micropipette, volume distribution of droplets became smaller than the previous results. Also the effect of dust particles within the micropipette on the volume of droplets generated was examined.
