Single-molecule fluorescence detection:its principle and demonstration

Abstract(in English) Single-molecule fluorescence detection was demonstrated with an optical microscope equipped with a position-sensitive photon- counting camera.Samples were fluorescent dyes adsorbed on a solid substrate.The photon-counting technique enables us to count fluorescence photons one by one.The number of fluorescence photons, which is theoretically evaluated,from single dye molecules coincides with that of being experimentally measured.The characteristics of the present technique are as follows:(1) identification of the position of single molecules within the limitation of the resolution of objective lenses,(2) simultaneous observation of the emitting spots including single molecules without scanning techniques,and (3) without requirement of low temperatures and high vacuums as sample preparations.
