Screening and application of enzymes suitable for biosensor component

Abstract(in English) The screening for bacterial enzymes for the biosensor has been carried outt. We isolated PQQ glucose dehydrognase from marine bacteria, strain N11. It has NaCl dependent activity, which was unique for marine bacterial enzyme. Two glucose dehydrogenases which could oxidize C-3 hydroxy group of glucose were found in marine bacteria. One was isolated from membrane fraction of Cytophaga marinoflava and the other was from soluble fraction of α-15strain. Using these enzymes, we developped an enzyme sensor and a novel fluorescent measurement method for 1,5-anhydroglucitol, diabetes marker in human serum. Strain SM4 was isolated from a soil near the hot spring. Its GDH showed unique temperature profile and thermal stability. Microbial sensor was constructed by using 1,5-anhydroglucitol assimilating bacterium. PQQGDH-overexpression changed substrate specificity of E. coli's respiratory activity.
