Abstract(in English) | The immobilization of antibody molecules is a key step in fabrication of immunosensors. Recently, an anti-2-phenyloxazollone lipid-tagged single chain antibody was biosynthetically produced by fusion with the signal peptide of the major lipoprotein of E. coli. We have investigated the immobilization of such antibody using the Langmuir Blodgett (LB) technique because it can control molecular orientation in monolayer level. Orientation and distribution of antibody in monolayer and kinetics of antigen binding are studied by using florescence microscopy and surface plasmon resonance (SPR). A stable mixed monolayer was formed by direct spreading of liposomes containing this fusion antibody. The transferred monolayer shows antigen binding activity on SPR response. |
