High efficiency transformation of fission yeast treated with thiol compounds by electroporation

Abstract(in English) a high efficiency method of Schizosaccharomyces pombe transformation by electroporation has been developed. In pretreatment with thiol compounds before the pulse application, the cells grown on a synthetic medium (SD) instead of a natural medium (YPD) drastically improved a transformation efficiency. The transformation efficiency of this protocol was approximately 10-fold higher than that of conventional treatment without Dithiothreitol. The best efficiency, approximately 1.2×10^7 transformants per μg of DNA, was achieved in the case of electric field 11kV/cm and its time constant 5 msec.
