Glutamate-induced neurotoxicity in neuron-astrocyte co-cultures from the embryonic rat cortex

Abstract(in English) Exposure of neuron-astrocyte co-cultures from the embryonic rat cortex to glutamate (Glu) resulted in neuronal death which was preceded by a long-lasting increase in intracellular Ca^<2+> concentration. Administration of (±)2-Amino-5-phosphonopentamicacid(AP5) protected the cells against neuronal death caused by Glu, suggesting that Glu-induced neuronal death and the long-lasting increase in intracellular Ca^<2+> concentration seem to be mediated by the activation of N-methy-D-aspartate(NMDA) receptors. Administration of dihydrokainate(DHK), an artrocytes'Glu-uptake inhibitor, resulted in exaggerated neuronal death even in the physiological level of extracellular Glu concentration. This result suggests that the Glu-transporter of astrocytes plays an important functional role in the protection of neurons against Glu toxicity.
