| 抄録(和) | これまで, RLGS(Restriction Landmark Genomic Scanning)法による2次元電気泳動像の解析システムの開発をしてきた. RLGS法は,2,3種類の制限酵素で切断したゲノムDNA断片を直接末端認識して2次元電気泳動によって展開・分画するものである。次のステージで遺伝子診断を目的として,すなわち,1つは,がん細胞から,他の1つは,正常なものから,臨床医が2つに分けたRLGS画像の集合から共通のパターンを自動的に生成する方法を提案した.この共通のパターンは,マスターパターンといい,すべてのスポットの座標や濃度から生成される.マスターパターンは,様々な疾病に特異的であったり,ある遺伝子の発現によるので,遺伝子診断の強力な道具とし使うことができる。 |
| 抄録(英) | A pattern analysis system of two-dimensional gel electrophoresis images based on RLGS (Restriction Landmark Genomic Scanning) method has been developed. It employs direct end-labeling of the genomic DNA fragments cut by two or three kinds of restriction enzyme and, separates them by means of 2-D gel electrophoresis. For the purpose of gene diagnosis in the next stage, we propose an automated synthesis method of the common pattern of multiple RLGS profiles for two sets; one comes from cancer cells, the other from normal, these are already classified by a clinician. The common pattern, which we call it 'master pattern', is generated by using the coordinates and density of all spots. As the master pattern is characteristic to a variety of diseases or depends on some gene expression, we can apply it to a powerful tools for gene diagnosis. |
