An in vitro Study of Cholonergic Modulation in the Rat Striatum

Abstract(in English) This study aims to elucidate actions of acetylcholine on corticostriatal synaptic transmission and the membrane excitability of striatal cholinergic interneurons. Whole-cell recordings were obtained from cholinergic interneurons in slice preparations of rats (postnatal day 14-23). Stimulation of the subcortical white matter evoked excitatory postsynaptic potentials (EPSPs) in cholinergic interneurons. Bath application of a muscarinic recepter agonist caused a slow depolarization and reduced the EPSP amplitude. This depolarization was antagonized by tetrodotoxin, a selective blocker of Na^+ channels. These data suggest that acetylcholine in the striatum promotes the membrane excitability of cholinergic interneurons indirectly and inhibits inputs from cortex to cholinergic interneurons.
