Abstract(in English) | Background: Nitric oxide (NO) and endothelium-derived hyperpolarizing factor (EDHF) size-dependently modulate vascular tone of coronary artery. We visualized in vivo both arterial and venous microcirculation in rats to examine whether hierarchical flow regulation of coronary vein depended on NO and EDHF. Methods: Subepicardial microvessels were observed by intravital videomicroscope under cyclooxygenase blockade. Coronary vascular responses to acetylcholine (ACh) were examined under two conditions; ACh and ACh + NO synthase inhibitor. We classified microvessels in terms of their diameters; arterioles & venules (<100μm), and small arteries & veins (>1OO μm). Results: Both arterioles and venules were dilated by ACh more than small arteries and veins, respectively, with greater dilation in arterial system. Small veins changed little after each intervention, while small arteries exhibited significant dilation. Both arterioles and small arteries were suppressed by NO synthase inhibitor, with greater residual dilation in arterioles. In venules, vasodilation was suppressed by NO synthase inhibitor like arterial side. Small veins did not response to NO synthase inhibitor. Conclusions: Coronary arterial and venous microvessels were hierarchically modulated by both NO and EDHF without any vascular response to these substance in small veins. |
