Abstract(in English) To investigate the possibility that tumor cells migrate toward intratumor blood vessels according to the gradient of oxygen concentration in tissue, we devised the gap cover glass (GCG) that allows to produce gradients of oxygen in the monolayer cultured cells. To verify the oxygen gradients, we conducted oxygen imaging in the extracellular medium in cultured MDA-MB-231 and COS-7 cells using a ruthenium complex under conditions of different cell density, cell species, or cellular respiration, all of which significantly alter the oxygen consumption rate per unit volume of the cell. The measured oxygen gradients were consistent with the changes in the oxygen consumption rate. Thus, we conclude that the GCG produces gradients of oxygen in monolayer cultured cells.
