Investigation of irradiation condition for real-time monitoring of sonoporation, using experimental design

Abstract(in English) In order to investigate the gene incorporation into cell by sonoporation using the optical microscope, the irradiation conditions of ultrsound were considered. The cultured cells (HepG2) with plasmid DNA (pEGFP-N3) and microbubbles (sonazoid), were exposure to the ultrasound. And the fluorescence intensity from Green Fluorescent Protein which was developed was measured. We assigned the following four factors to L9-orthogonal table of an experimental design and analyzed the factors which induce large the changes of the fluorescence intensity. [ (1) distance between ultrasound transmitter and bottom of microplate where the cells are cultured (2) intensity of exposing ultrasound, (3) exposing period, (4) duty ratio of exposing ultrasound ] The intensity of fluorescence of the GFP was shown to be influenced by the distance and the intensity of ultrasound.
