Abstract(in English) The high-resolution imaging of fine structures such as neurites requires objectives with high numerical apertures (NAs). However, spherical aberration (SA) due to the mismatch of refractive indices between an immersion medium and samples, often distorts acquired images. Whereas a correction collar attached to high NAs objectives has been designed to compensate SA, its manual adjustment and acquisition of the best image of a given samples is the most difficult mission. To solve the problem, we have developed SA compensation system, by motorizing collection collar and formulating an algorithm which calculates the optimum collection collar position based on contrasts of images. This system allows quantitative analysis of fine structures of cerebral cortex in vivo. Furthermore, as this system enables us to estimate the refractive index of a given samples on the basis of the compensation degree, we found the cortical structure could be optically heterogeneous along the z-axis in the living brain.
