Effect of reaction conditions on magnetic signal from magnetic markers in liquid-phase immunoassays

Abstract(in English) We have been developing a highly sensitive liquid-phase detection of biological targets using magnetic markers and HTS SQUID. In order to realize high sensitivity of this technique, it is necessary to decrease signal from free markers, which we call blank signal, as well as to increase signal from bound markers. Previously, we showed that dispersion process after magnetization is effective to decrease blank signal. In this study, we optimized reaction conditions when the dispersion process was introduced, i.e., density of magnetic markers and time used for reaction. We also measured diameter of magnetic markers before and after the magnetization so as to study the aggregation caused by the magnetic field. It was found that magnetic field accelerated aggregation of magnetic markers. Then, we calculated blank signal from the agglomerate of free markers by taking into account their Brownian relaxation. The calculated blank signal reasonably agreed with the experimental result. Finally, we measured the Brownian relaxation time of the bound markers, and found that the measured time was shorter than the theoretical expectation.
