Construction of a cost-efficient confocal laser-scanning microscope

Abstract(in English) Confocal laser-scanning microscopes (CLSMs), which enable to obtain three-dimensional image with high resolution cannot be achieved in conventional optical microscope, have been widely utilized in various fields such as, materials science, semiconductor and electronic industries, biological and medical science, and so on. However, such CLSMs are usually quite expensive. In this study, we have constructed cost-efficient CLSM system combining various components with low cost. We have employed galvano mirror pair to scan focal spot of the laser light in two-dimensional horizontal plane, and piezo device to which an objective lens can be attached to scan along optical (vertical) axis, respectively. We used MPPC (Multi-Pixel Photon Counter) module for laser intensity measurement. For automated scanning of laser spot and recording signals, we used LabVIEW. By employing low-cost optical components, we have constructed cost-efficient CLSM system at an expenses of ~ 3,000,000.
