Influence estimation of the process for histology on the measurement of acoustic properties using high frequency

Abstract(in English) In bioacoustics microscopy, the effect of tissue preparation, e.g. formalin fixation, on acoustic characteristic of sectioned samples with thickness of 4 ~ 12 µm is unknown. This study aims to estimate the influence of duration of formalin fixation on speed of sound (SOS) of normal rats liver. In addition, focusing on the difference between methods with or without formalin fixation, we compared SOS between tissue made by paraffin embedded method and frozen method. As a result, SOS increased up to 1650-1750 m/s from 1610-1680 m/s during 0-6 hours of duration of formalin fixation. The comparison of paraffin embedded method with frozen method demonstrated that the difference in SOS between these methods were 50-100 m/s. SOS of sample in frozen method was approximately consistent with SOS of the liver just after the harvest in a previous study.
