Abstract(in English) | By using affinity capillary electrophoresis, binding equilibrium of an arylsulfonamide (SA) to a series of metal-substituted carbonic anhydrase (M-CA) and kinetic stability of M-CA toward solvolysis were studied. In the binding of SA, the hydrophobic cavity of CA plays an important role to force the SA to donate the amide moiety to the metal center of CA to fulfill the tetrahedral coordination geometry. In the kinetics, the hydrophobic cavity excludes the access of EDTA to the metal center, while the tetrahedral coordination geometry supported by three histidine residues gave the Zn(II) center exceptionally high kinetic stability. |
