Abstract(in English) | Blood coagulation tests are in increasing demand as a preoperative test to evaluate hemostatic function. We have been developing a method to evaluate blood coagulation by detecting the changing signal of a mixture of magnetic nanoparticles (MNPs) and blood using the developed HTS-SQUID AC magnetic measurement system, taking advantage of increasing the viscosity of the blood due to clotting. First, we measured the time evolution of the magnetic signal of MNPs in the CaCl2 solution because the CaCl2 solution is mixed with blood in a blood coagulation test to accelerate clotting. As a result, the magnetic signal decreased to 18% of that in pure water within 2 minutes of mixing due to the aggregation of MNPs induced by Ca2+ ions. Next, we measured the time evolution of the magnetic signal in the case of mixing with MNPs and sheep plasma. Without adding the CaCl2 solution, the time evolution of the magnetic signal could be fitted by the single exponential function with a time constant of 10.0±0.603 min due to aggregation. On the other hand, adding the CaCl2 solution, the time evolution of the magnetic signal was fitted by the exponential double function due to aggregation and clotting, with a time constant of 1.57±0.862 min and 12.9±1.70 min. Therefore, we suggested that the blood coagulation could be evaluated by measuring the time evolution of the magnetic signal. |
