Abstract(in English) | DOX-Alb is albumin nanoparticles of approximately 160 nm in diameter with anticancer drug doxorubicin (DOX) adhering to their surfaces. DOX-Alb is used to deliver the drug to tumor tissue by intravascular injection of DOX-Alb with microbubbles and sonication. This study developed a capillary phantom with a 10 ?m diameter lumen using acrylamide gel to validate the basic idea method. An experiment to introduce DOX or DOX-Alb into the lumen confirmed that the phantom has the characteristics of diffusing low molecular weight DOX and retaining DOX-Alb nanoparticles inside the lumen, as in biological capillaries. Another experiment to irradiate the phantom by one-shot ultrasound burst pulse (1 MHz, 1 MPa in negative peat pressure, and 50 or 1,000 cycles) showed that bubble oscillation enhances DOX diffusion and induces lumen deformation and leakage of DOX-Alb. This phantom is expected to play an essential role in understanding the phenomena occurring in capillaries. |
