Low-cost fabrication of structured illumination microscope using 3D-printed flexure stage

Abstract(in English) The OpenFlexure stage and microscope, which is open source and can be replicated by 3D printing, uses a flexure mechanism to achieve extremely high positioning control with submicron precision. However, it is an ordinary optical microscope and does not have depth resolution. In this study, we worked on the improvement of the microscope to a structured illumination microscope with optical sectioning capability by adding printed parts for structured illumination. The additional parts were designed using an open CAD software, OpenSCAD, and were fabricated by 3D printing. The stage motion control, image acquisition, and analysis can be performed on a Raspberry Pi using Python. The Raspberry Pi can be remotely controlled from a PC via Wi-Fi. Optical sectioning images of fibers in a nonwoven mask were obtained using a structured illumination microscope.
