Abstract(in English) Once a balance between excitation and inhibition in a brain is disrupted, mental disorder will occur. One of the disorders is the temporal lobe epilepsy. Fifteen percent of the epileptic patients cannot get over with the medicine. θ rhythm suppresses the epileptic discharges. In the previous study we found that β rhythm also will suppress the generation of the discharges, and propose the stimulation with β frequency is a therapy for the epileptic patients. However, it is not yet found whether the suppression of the epileptic discharges by β rhythm occurs in vivo. We made kainate-induced epileptic model of rat, and have been studying the effect of β rhythm on the epileptic discharges. In the present study, we study the hippocampal neuronal network in the rat model. The rats in kainate (KA) group had the significantly more frequent Grooming behavior a week after KA injection than those in Control group. The rate of spontaneous activity was higher in hippocampal slices in KA group than in Control one. The slices in KA group also had the spontaneous activities with longer duration. There were the spontaneous activities observed in KA slices and not seen in the Control slices. When a cholinergic agent carbachol was applied to the slices, the activities were diminished in both groups, and carbachol-induced β oscillations (CIBO) were induced. The frequencies of CIBO in KA were significantly higher than those in Control. In addition, the application of carbachol with picrotoxin GABAA receptor antagonist decreased CIBO frequency, which was about the same as that in Control. These results suggest that the inhibitory neurons are more activated in hippocampus in KA group, β wave will suppress the spontaneous activities related to the epileptic discharges.
