3D cell culture in silica nonwoven fabrics

(in English) In this study, we examined silica nonwoven fabrics (SNF) as scaffolds of mesenchymal stem cells (MSCs) for bone and cartilage regeneration and fibroblast for reconstituted human skin. Human bone marrow-derived MSCs seeded in SNF proliferated and thickness of cell layers became over 80 m after 14-days culture. SNF enhanced osteogenic differentiation of MSCs comparing to 2D culture on tissue culture-treated polystyrene. The level of chondrogenesis observed in MSCs seeded within was comparable to that in atelocollagen gels, which are widely used for chondrogenesis of MSCs in vitro and in vivo. Fibroblast also migrated in the SNF and cellular layer of fibroblast were formed. These results indicated the availability of SNF as scaffolds of MSCs for osteo-chondro regeneration and fibroblast for reconstituted human skin.
