Construction of an artificial cell membrane device and its application to a sensor for drug side effects

(in English) Ion channel proteins are membrane proteins that regulate ion flows across cell membranes and have attracted attention not only as primary molecular targets for drug actions but also as major targets for drug-induced side effects. Reconstitution of ion channel proteins in artificial bilayer lipid membranes (BLMs) provides an excellent system for drug screenings. The advantage of this system is its versatility in terms of producing target channels, including those prepared by cell-free protein expression systems, in which pure proteins of various channel genotypes can be produced directly from a protein-encoding plasmid. However, the BLM system suffers from low experimental efficiency due to instability of the BLMs and difficulty in ion channel incorporation into BLMs. In this presentation, we will show you our recent approaches for addressing these issues based on nano- and micro-fabrication. Our approach leads to the realization of a BLM-based high-throughput platform for screening the effects of drugs that act on various ion channel genotypes.
