[Poster Presentation] Effect of sonication frequency on inactivation of bacteria and fungi

(in English) Heat, ultraviolet light, and chemicals have all been used for disinfection of foods, but unpleasant tastes and denaturation are often cited as problems with these methods. Disinfection by ultrasonic waves (sonication) has attracted attention because it has less influence on food quality. Previous studies have shown that ultrasonic cavitation causes bacterial and fungal inactivation by both physical and chemical mechanisms. However, determining the mechanism of inactivation can be complicated, as it differs depending on the nature of the microorganisms. In this study, the characteristics of bacterial and fungal inactivation via different frequencies of ultrasonic waves were investigated in the fungus Saccharomyces cerevisiae, and in the bacteria Bacillus subtilis and Escherichia coli. From these experiments it was found that the optimum frequency for inactivation varies depending on the nature of the microorganisms. Presumably, the deactivation mechanism is different at different frequencies, but is likely to involve, among other things, active oxygen, cellular destruction, and cellular mechanical resonance. Furthermore, we examined the effect of acoustic power on E. coli at the optimum inactivation frequency.
