Basic study on usefulness of electrically induced microbubbles for in vitro sonoporation

(in English) We have been studying a sonoporation technique that uses pulsed ultrasound and microbubbles for gene or drug transduction into cells. In vitro sonoporation experiments, monolayer cells cultured on a coverslip were dipped into microbubbles suspension to attach the bubbles on the cells, and then the cells were irradiated by ultrasound to induce cell membrane perforation. Microbubbles of an ultrasound contrast agent are widely used for sonoporation studies, but it is difficult to control sizes and locations of bubbles that attaches to cells. In this study, we investigated a technique to generate microbubbles with a small variation in diameter and with several types of shell material by electrically induced. In vitro sonoporation experiment was carried out, indicating that the bubbles can induce sonoporation effect of cultured cells.
