Highly Accurate Reconstruction of Microscope 3D images based on Micro Anatomic Structures

(in English) The objective of this research is to develop a method for accurately
registering 2D microscope images of spatially successive slices of an
organ. One can obtain a 3D microscope image from the 2D slice ones by
stacking them.The proposed method registers given microscope images
based on the smoothness of the anatomical structures in the images.
One needs to non-rigidly register the 2D images before stacking them
because the slices are non-rigidly deformed when they are mounted on
slide glasses.
The proposed method firstly extracts the cross sections of the
anatomical structures such as the micro blood vessels from each of the
given microscope images, then makes correspondences between the
extracted cross sections among the images, and finally deforms each
image so that the anatomical structures have smooth surfaces in the
resultant reconstructed 3D image.
The authors reports experimental
results obtained by applying the proposed method to a set of
microscope images of the pancreases of KPC mice.
