[Invited Talk] Non-invasive analysis of the whole brain neuronal activity using MRI -- from fundamental to application --

(in English) Recently, visualization of the dynamics of intracellular Ca^{2+} increased by the neuronal action potentials (Ca^{2+} imaging) has been widely used for raveling the functions of the nervous systems. The Ca^{2+} imaging can record the multicellular activities simultaneously, however, it can hardly visualize the whole brain activities. Thus, for visualizing the whole brain activities, we used one MRI technique, activation-induced manganese-enhanced MRI (AIM-MRI), which is based on the use of Mn^{2+} as a surrogate marker of Ca^{2+} influx. Therefore, it is known that AIM-MRI can measure the history of the neuronal activities in whole brain. After confirmation of the dependency of Mn^{2+} accumulation to neuronal activity, we demonstrated that AIM-MRI with quantitative determination of the longitudinal relaxation time (qAIM-MRI) reveals the severity of Parkinson’s disease (PD) in mice. A highly active region was then observed by qAIM-MRI in the caudate-putamen in PD-model mice that was significantly correlated to the severity of PD, suggesting its involvement in the expression of PD symptoms. Our results provide a foundation for the development and application of quantitative activity mapping in the entire brain using qAIM-MRI for the study and diagnosis of various neurological disorders.
