[Poster Presentation] Effect of Ghrelin on hypothalamic tuberomammillary nucleus neurons

(in English) Ghrelin, which is produced in the brain and stomach, is an endogenous ligand of the growth hormone secretagogue receptor (GHS-R). Ghrelin participates in regulation of GH release and food intake. Recent study demonstrated that ghrelin also participates in regulation of sleep-wakefulness. Tuberomammillary nucleus (TMN), which is one of brain sites that control sleep-wakefulness, also expresses GHS-R. However, effect of ghrelin on TMN neurons is remained unclear. Thus, we examined effects of ghrelin on TMN neurons using rat brain slice preparations and whole-cell patch clamp recording technique. Application of ghrelin depolarized TMN neurons and ghrelin-induced depolarization was also observed under presence of tetrodotoxin. These results suggest that ghrelin depolarizes TMN neurons and participates in the regulation of sleep-wakefulness by stimulated activity of TMN neurons.
