The structure, dynamics and hydration behavior of collagen model polypeptides, (Pro-Pro-Gly)n (PPGn), were investigated in aqueous media over a wide temperature range using broad band dielectric relaxation (DR) techniques that spanned frequencies from 1 kHz to 20 GHz. PPG5 molecules dissolved in solution as single chains without forming triple helices due to their low molecular weight or length, and behaved random coils bearing fully ionized amino and carboxy termini and the hydration number ca. 9 per amino acid residue. On the other hand, PPG10 formed mixtures of mono-disperse rods as triple helices and random coils of single chains. Since the fractions of these states were precisely determined via the obtained DR spectra, the triple helix-to-single coil transition was considered thermodynamically based on the equilibrium constant calculated from the fractions assuming the triple helix-to-single coil transition as a kind of chemical reaction. Furthermore, the hydration number of PPG10 in the triple helix state in solution was estimated to be ca. 2 per amino acid residue and was close to the number of water molecules hydrated to PPG10 in single crystals forming triple helices. |
