アルツハイマー型認知症は,アミロイドβやアミロイドβに関連した物質の変化に注目して研究が進められてきたが,系全体の変化に注目された研究は少なく,メカニズムの解明にはいたっていない.本研究はアルツハイマー発症時に影響を受ける海馬シグナル伝達系における構造的特徴,およびそれを規定するメカニズムの解明を目標とした.重み付き有向グラフで表現されたシグナル伝達経路と遺伝子発現データを考慮してアルツハイマー発症時のネットワークを作成し,従来の統計的な指標とk最短経路による解析を行った.その結果,ネットワークの密度、推移性、平均クラスター係数、最短経路長といった従来の統計的な指標は似ているにもかかわらず,k最短経路による解析で得られた特定の入出力における信号伝達の効率が異なっていた.本モデルにより,遺伝子調節を用いてアルツハイマー発症時に起こる海馬シグナル伝達の頑強性の変化を示すことに成功した. |

Alzheimer's disease (AD) has been studied for the changes of molecules such as amyloid β. Few studies have investigated the change of the whole signal transduction system, preventing the elucidation of the mechanism. We aimed to elucidate the structural features of human hippocampal signal transduction system and the rules and mechanisms which emerge in AD. We constructed AD specific signal transduction network as a graph using gene expression and weighted directed signal transduction pathway data of hippocampus, and evaluated the changes of the system by commonly used statistical indexes and k shortest path. As a result, although the network density, transitivity, average clustering coefficient and shortest path length in the AD network resembled those in the network, the nodes of which were removed randomly, the robustness differed in the specific input and output sets in the k shortest path analysis. Applying this model, we successfully identified the changes of robustness in human hippocampal signal transductions in AD due to the differential gene expressions. |
