The characteristics of endothelial cell-smooth muscle cell cocultures in LDL-uptake and cell proliferation

(in English) An attempt was made to construct an EC-SMC coculture as a model of an arterial wall by seeding bovine arterial endothelial cells (ECs) directly over smooth muscle cells (SMCs) and coculturing them. It was found that to construct an EC-SMC coculture in which ECs are in direct contact with SMCs and covering them as a confluent monolayer, it is necessary to have ascorbic acid at a concentration higher than 50 mg/mL. It was also found that both the uptake of LDL by the cells in the EC-SMC coculture and the thickness of the cell layer of the EC-SMC coculture increased with increasing the concentration of LDL in a culture medium.
