膵β細胞は、インスリン分泌と関連して膜電位の変化を起こす。通常の膵β細胞では、膜電位の変化において周期的なバースト波を示すが、糖尿病患者などのインスリン分泌に障害が生じている膵β細胞では、バースト波が非周期的となる。本研究では、膜電位の変化を数理モデルで再現し、そのモデルに対するノイズの影響について検討した。その結果、ノイズを加えることにより、インスリン分泌を意味するスパイク数が増加することを確認した。また、ノイズ強度を大きくするほどスパイク数の増加がより大きくなる傾向にあることも確認した。さらに、周期的なバースト波を示すパラメータよりも非周期的なバースト波を示すパラメータにおいて、ノイズの影響によるスパイク数の増加が大きい傾向にあることをシミュレーションにより示した。これは、インスリン分泌に障害がある膵β細胞に対しては、障害のない場合に比べて、ノイズ刺激がよりインスリン分泌を促進する可能性を示唆しており、糖尿病治療等に貢献する可能性がある。 |

The membrane potential of pancreatic beta cells changes its value in accordance with insulin secretion. In normal beta cells, the change of membrane potential shows periodic bursting, while the bursting of cells in diabetes patients is not periodic, namely the insulin secretion is disordered. In this study, the dynamics of the membrane potential is reproduced by numerical simulation of a mathematical model, and the effect of noise on the model is investigated.
As a result of the simulation, we confirm that the number of spikes which corresponds to insulin secretion increases due to noise. We also confirm that the stronger the noise amplitude is, the more the number of spikes increases.
Furthermore, the number of spikes more increases for the parameters of the model in which the bursting is not periodic than for that of periodic bursting. This result implies that noise stimulation can promote the amount of insulin secretion for the disordered beta cell compared with the normal cell. Thus, the noisy stimulation would be useful for the remedy for diabetes. |
